NF-κB-GFP報告基因是自主研發(fā)的用于檢測NFκB轉(zhuǎn)錄活性水平為目的的報告基因。 NF-κB基因在炎癥反應(yīng)、**應(yīng)答和抑制細(xì)胞凋亡等方面有著重要的作用。
NF-κB-GFP報告基因主要應(yīng)用于NF-κB信號通路的研究和基于NF-κB信號通路的**研究以及癌細(xì)胞中NF-KB的調(diào)控機(jī)制的研究。
pGMNF-ΚB-GFP是改造后的哺乳動物真核表達(dá)載體,在其多克隆位點(diǎn)插入了多個NF-κB結(jié)合位點(diǎn),可以高效地檢測NF-κB的激活水平。由于載體采用了GFP作為報告基因,更便于后續(xù)的檢測。同時,對載體中預(yù)測出的其它轉(zhuǎn)錄因子以外的結(jié)合位點(diǎn)進(jìn)行了適當(dāng)?shù)耐蛔?,增加了質(zhì)粒的轉(zhuǎn)錄因子結(jié)合特異性。另外,由于質(zhì)粒體積減小,使得NF-κB-GFP報告基因更易于轉(zhuǎn)染。NF-κB GFP Reporter Plasmid(NF-κB-GFP報告基因質(zhì)粒)

使用說明
pGMNF-κB -GFP可以采用常規(guī)轉(zhuǎn)染方法轉(zhuǎn)染哺乳動物細(xì)胞。
TNF-α、IL-1β和LPS等是常見的NF-κB激活劑,可作為NF-κB報告基因的陽性對照。
