STAT3-luc報告基因慢病毒是翊圣生物自主研發(fā)的用于檢測STAT3轉錄活性水平為目的的報告基因慢病毒顆粒。STAT3(signal transducers and activators of transduction-3)報告基因與多種惡性腫瘤的發(fā)生、發(fā)展和侵襲轉移關系密切。
STAT3報告基因慢病毒主要用于監(jiān)控細胞內的STAT3轉錄活性、腫瘤****的研發(fā)以及基因過表達和RNAi的表型分析等。
pGMLV-STAT3-Lu是在pGMLV-Lu慢病毒載體的多克隆位點插入了多個STAT3結合位點,可以高靈敏度地檢測STAT3的激活水平。采用慢病毒作為報告基因的優(yōu)點在于它能夠感染多種難感染的細胞,比如原代細胞、干細胞、神經元細胞等。而且慢病毒能夠將外源基因插入細胞基因組內,實現穩(wěn)定轉染。Lentivirus STAT3 Luciferase Reporter STAT3-Luc 報告基因慢病毒顆粒

使用說明
STAT3報告基因慢病毒顆粒采用慢病毒轉染方法轉染哺乳動物細胞。用熒光素酶檢測試劑盒或雙熒光素酶檢測試劑盒進行檢測。IL-6、白血病抑制因子(LIF)和血小板生長因子(PDGF)等可激活STAT3,可作為STAT3報告基因的陽性對照。
注意事項
1)病毒操作時*好使用生物**柜,如使用普通超凈工作臺操作病毒,請不要打開風機。
2)病毒操作時請穿實驗服,帶口罩和乳膠手套。
3)操作病毒時必須特別小心,不要產生氣霧或飛濺。如操作時超凈臺有病毒污染,立即用10%次氯酸鈉溶液搽拭干凈。接觸過病毒的槍頭、離心管和培養(yǎng)板等需用10%次氯酸鈉溶液浸泡1h以上后棄去。
4)用顯微鏡觀察細胞感染情況時應遵從以下步驟:擰緊培養(yǎng)瓶或蓋緊培養(yǎng)板。用70%乙醇清理培養(yǎng)瓶外壁后到顯微鏡處觀察拍照。離開顯微鏡試驗臺前,用70%乙醇清理實驗臺。
5)病毒操作完成后,用肥皂清洗雙手。
