GAL4熒光素酶報(bào)告基因（報(bào)告基因質(zhì)粒）（GAL4 luciferase reporter plasimd）是自主研發(fā)的用于檢測GAL4轉(zhuǎn)錄活性水平為目的的報(bào)告基因。GAL4是酵母（Saccharomyces cerevisiae）轉(zhuǎn)錄激活蛋白，是酵母半乳糖的代謝相關(guān)基因。

GAL4報(bào)告基因主要用于檢測蛋白質(zhì)相互作用的酵母雙雜交系統(tǒng)，以及在其基礎(chǔ)上發(fā)展出來的用于檢測Protein-DNA相互作用的酵母單雜交系統(tǒng)和酵母三雜交系統(tǒng)。

pGMGAL4-Lu是改造后的哺乳動(dòng)物真核表達(dá)載體，在其多克隆位點(diǎn)插入了多個(gè)GAL4結(jié)合位點(diǎn)，可以高靈敏度地檢測GAL4的激活水平。同時(shí)，對載體中預(yù)測出的其它轉(zhuǎn)錄因子以外的結(jié)合位點(diǎn)進(jìn)行了適當(dāng)?shù)耐蛔?，增加了質(zhì)粒的轉(zhuǎn)錄因子結(jié)合特異性。由于質(zhì)粒體積減小，使得GAL4報(bào)告基因質(zhì)粒更易于轉(zhuǎn)染。GAL4 luciferase reporter plasmid （GAL4-Luc熒光素酶報(bào)告基因質(zhì)粒）

使用說明

pGMGAL4-Lu可以采用常規(guī)轉(zhuǎn)染方法轉(zhuǎn)染哺乳動(dòng)物細(xì)胞。用熒光素酶檢測試劑盒或雙熒光素酶檢測試劑盒進(jìn)行檢測。
