C/EBPa熒光素酶報(bào)告基因(報(bào)告基因質(zhì)粒)(C/EBPa luciferase reporter plasimd)是自主研發(fā)的用于檢測(cè)C/EBPa轉(zhuǎn)錄活性水平為目的的報(bào)告基因。C/EBP(CCAAT enhancer binding proteins)是一組耐熱的轉(zhuǎn)錄因子,主要對(duì)調(diào)控有絲分裂生長(zhǎng)停滯和分化。
C/EBPa報(bào)告基因主要檢測(cè)細(xì)胞中C/EBPa調(diào)節(jié)信號(hào)通路的活性、**研究以及基因過(guò)表達(dá)和RNAi的表型分析等。
pGMC/EBPa-Lu是改造后的哺乳動(dòng)物真核表達(dá)載體,在其多克隆位點(diǎn)插入了多個(gè)C/EBPa結(jié)合位點(diǎn),可以高靈敏度地檢測(cè)C/EBPa的激活水平。同時(shí),對(duì)載體中預(yù)測(cè)出的其它轉(zhuǎn)錄因子以外的結(jié)合位點(diǎn)進(jìn)行了適當(dāng)?shù)耐蛔?,增加了質(zhì)粒的轉(zhuǎn)錄因子結(jié)合特異性。由于質(zhì)粒體積減小,使得C/EBPa報(bào)告基因質(zhì)粒更易于轉(zhuǎn)染。C/EBPa luciferase reporter plasmid (C/EBPa--Luc熒光素酶報(bào)告基因質(zhì)粒)
