Age: M. dunni細(xì)胞*****
Gender: female
Comments: Clone IIIC8 was derived from tail skin of a normal female Mus dunni mouse.
This line supports the replication of all four classes of MuLV, i.e., ecotropic, xenotropic, amphotropic and mink cell focus forming (MCF).
The cells exhibit cytopathic effects when infected with most ecotropic and MCF viruses.
They are resistant to infection by Moloney MuLV (presumably due to post translational modifications of the receptor), and lack most endogenous MuLV sequences.
To maintain the cells as confluent cultures (in viral assays, for example), use RPMI 1640 or Dulbecco's modified Eagle's medium.
Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated McCoy's 5a Medium Modified, Catalog No. 30-2007. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Atmosphere: air, 95%; carbon dioxide (CO2), 5%
Temperature: M. dunni細(xì)胞37.0°C
Subculturing: Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:8 is recommended
Medium Renewal: Twice per week
Remove spent medium, add fresh 0.25% trypsin, 0.03% EDTA solution, rinse and remove.
Add another 1 to 2 ml of fresh trypsin solution, and allow the cells to sit at 37C (or at room temperature) until the cells detach.
Add fresh medium, aspirate and dispense into new flasks.
Preservation: Freeze medium: Complete growth medium, 95%; DMSO, 5%
Storage temperature: liquid nitrogen vapor phase
