Cytogenetic Analysis: This is a near-diploid cell line of male origin with a modal chromosome number of 45 and a polyploidy rate of 10%. The karyotype is highly rearranged with most of the cells containing the t(11;14)(q13;q32) common to mantle cell lymphoma. Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing: Protocol: Cultures can be maintained by the addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 1 X 10(5) viable cells/ml. Maintain cultures at a cell concentration between 3 X 10(5) and 1 X 10(6) cells/ml. Do not allow the cell concentration to exceed 1 x 10(6) cells/ml.
Subcultivation ratio: A subcultivation ratio of 1:3 to 1:6 is recommended.
Interval: Subculture when cell concentration is between 3x10(5) and 1x10(6) cells/ml.
Medium renewal: Add fresh medium every 2 to 3 days (depending on cell density).
