Comments: A7細(xì)胞M2 cells are severely deficient in filamin-1 mRNA and protein. Filamin-1, also known as actin-binding protein (ABP, APB-280), cross-links actin filaments in vitro and connects these filaments to plasma membrane glycoproteins. [42092]
The filament-1 gene on the X chromosome is present in these cells.
ABP (filamin-1) was transfected into M2 cells in a mammalian expression vector (LK444) containing full length human ABP recombinant DNA (LK-APB). [42092]
A7 cells express levels of filamin-1 comparable to wild-type filamin-1 containing melanoma cells. [42092]
Filamin-1 is essential for cell migration and has been shown to bind partners in cell signal transduction, cell membrane sorting and cytoskeleton-membrane association.
A7 cells are useful as a control for studying these systems.
Propagation: ATCC complete growth medium: Minimum essential medium (Eagle) with 2 mM L-glutamine and Earle's BSS adjusted to contain 1.5 g/L sodium bicarbonate, 0.1 mM non-essential amino acids, and 1.0 mM sodium pyruvate supplemented with 10 mM HEPES, 0.5 mg/ml G418 and 10% fetal bovine serum
Temperature: 37.0°C
Subculturing: A7細(xì)胞Protocol: Remove medium, and rinse with 0.25% trypsin - 0.53 mM EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach. Add fresh culture medium, aspirate and dispense into new culture flasks.
Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:8 is recommended
